Goal reports that the Frenchman sustained a knee injury in training on Tuesday and is set to miss the mammoth clash in Madrid later today, Wednesday, February 25.
The former Paris Saint-Germain forward was forced to leave training due to pain in his left leg.
Mbappé is currently the top scorer in the Champions League with 13 goals in eight appearances for Los Blancos.
The 27-year-old's absence will be a huge blow for Álvaro Arbeloa's side.
Real Madrid go into the game with a 1-0 advantage from the first leg in Portugal and are clear favourites to advance to the next stage of the competition.
