The former West Brom defender was booked for the second time in the qualifiers during Nigeria's 4-0 thrashing of Benin Republic last week.
However, reports indicate that Eric Chelle may still call up Ajayi for the mini playoff tournament due to his availability for the final, should Nigeria qualify to face DR Congo or Cameroon.
Ola Aina, who also sustained a hamstring injury against South Africa in September, remains sidelined and will miss the clash against Gabon.
New reports emerging indicate that Super Eagles attacking duo Cyriel Dessers and Terem Moffi have both sustained injuries.
According to Rotowire, Moffi has suffered a calf injury despite recently returning from a long-term layoff.
Cyriel Dessers is the latest player to join the injury list, following an ankle injury last weekend while playing for Panathinaikos.
The 30-year-old striker missed the October World Cup qualifiers after losing his spot to Sevilla forward Akor Adams.
